Protease Inhibitor Cocktail Tablets, EDTA-Free
Protease Inhibitor Cocktail Tablets, EDTA-free are designed to protect isolated proteins by providing an extraordinary inhibition of serine proteases, cysteine proteases and aminopeptidases during extractions from animal and plant tissues or cells, yeast and bacteria. The provided tablets do not contain EDTA, leaving the stability and the function of metal-dependant proteins unaffected. Protease Inhibitor Cocktail EDTA-free, Tablets contain both irreversible and reversible protease inhibitors. Aspartic and metalloproteases are not inhibited with the exception of aminopeptidases. The affinity purification of Poly-His tagged fusion proteins via IMAC (immobilized metal ion affinity chromatography) is also facilitated (no dialysis necessary).
Composition
The tablets contain the following protease inhibitors: AEBSF, Aprotinin, E-64 and Leupeptin Hemisulfate.
20 tablets provided in a glass vial. Each tablet is sufficient for the inhibition of the proteolytic activity in 50 ml extraction solution

Product Usage / Preparation
A 50X stock solution of one tablet has an adsorption of 0.35 at 280 nm.
The inhibitory activity of each lot is tested with exactly defined trypsin and papain assays at 37oC. The detection is performed with a universal protease substrate: resorufin-labeled casein. Typically, the proteolytic activity is inhibited by 90% after one hour.
・Working Solution
Protease Inhibitor Cocktail EDTA-free, Tablets directly in the extraction medium. Depending on the proteolytic activity, one tablet should be sufficient for the inhibition of the existing proteolytic activity in 50 ml. If very high proteolytic activity is present, one tablet should be used for 25 ml extraction buffer. If it is necessary to inhibit proteolytic activity in a smaller volume, we recommend using a stock solution (50X conc.).

Dissolve one tablet in 1 ml distilled water.
Storage and Handling
Store dry at 4oC, if store correctly, Protease Inhibitor Cocktail Tablets, EDTA-free are stable until the expiration date indicated on the vial. The stock solution is stable for 1-2 weeks, stored at 4oC.
